Меньщикова</copyright-holder></permissions><self-uri xlink:href="https://vietnamjournal.ru/0041-3771/article/view/669638">https://vietnamjournal.ru/0041-3771/article/view/669638</self-uri><abstract xml:lang="en"><p id="idm45181326114448">Despite the key role of the Keap1/Nrf2/ARE redox-sensitive signaling system in cellular metabolism, little is known about its relationship to lysosome biogenesis. In this paper, a theoretical and experimental analysis of the possibility of such a link has been carried out. By forming a position frequency matrix in the transcription factor genes TFEB and TFE3, the presence of a large number of ARE-like sequences was found in the non-coding regions. In vitro exposure to J774 cells by Keap1/Nrf2/ARE activators (original synthetic monophenol TS-13 and <italic>tert</italic>-butylhydroquinone as comparison compound) results in dose-dependent induction of <italic>Tfe3</italic> and <italic>Tfeb</italic> genes, accompanied by a gradual increase in the lysosome number and autosomal-lysosomal fusion intensity. Thus, it can be assumed that the proteins controlling the ARE-dependent genes are able to influence lysosome biogenesis.</p></abstract><trans-abstract xml:lang="ru"><p id="idm45181326113104">Несмотря на ключевую роль редокс-чувствительной сигнальной системы Keap1/Nrf2/ARE в клеточном метаболизме, практически ничего не известно о ее связи с биогенезом лизосом. В настоящей работе выполнен теоретический и экспериментальный анализ возможности наличия такой связи. Путем формирования позиционной матрицы в генах транскрипционных факторов TFEB и TFE3 обнаружено наличие в некодирующих участках большого количества ARE-подобных последовательностей. Воздействие <italic>in vitro</italic> на клетки J774 активаторами системы Keap1/Nrf2/ARE (оригинального синтетического монофенола ТС-13 и препарата сравнения трет-бутилгидрохинона) приводит к дозозависимой индукции генов <italic>Tfe3</italic> и <italic>Tfeb</italic>, сопровождающейся постепенным увеличением количества лизосом и интенсивности аутофагосомно-лизосомного слияния.
